ABSTRACT

Panax ginseng, a perennial herb, is prone to diseases and insect pests in the growth process, which are primarily prevented and treated by pesticides. However, due to the lack of standardization in the types, frequencies, and doses of pesticides, pesticide residues have become the main exogenous pollutants of P. ginseng. To explore the risk of pesticide residues in P. ginseng, this paper summarized and analyzed the common pesticide residues in P. ginseng, detection techniques, and pesticide residue limit stan-dards based on the published literature in recent years. The results revealed that the main pesticide residues in P. ginseng were organochlorine pesticides, such as tetrachloronitrobenzene, pentachloronitrobenzene, and hexachlorobenzene, and the detection techniques were dominated by gas chromatography(GC), liquid chromatography(LC), or those combined with mass spectrometry(MS). Because of the long half-life and difficulty in degradation, organochlorine pesticides have become the main factor affecting the export of P. ginseng. It is worth mentioning that P. ginseng has been classified as food in Japan, South Korea, the European Union, and other countries, and the standards of pesticide residues and limits are stricter than those in China. The quality and safety of P. ginseng are prerequisites for the efficacy of Chinese medicine and the development of traditional Chinese medicine. The formulation of scientific and effective standards for pesticide application and limits would promote the high-quality development of the P. ginseng industry.

ABSTRACT

The present study counted the frequency of detection technologies and monitoring frequency of pesticide species by frequency analysis based on the 28 258 pieces of data on pesticide content of Chinese medicinal materials in CNKI, calculated the detection rate and exceeding rate of different types of pesticides, and systematically analyzed the pesticide residue pollution of Chinese medicinal materials. The results showed that there were 40 types of pesticides with detection rates higher than 10%, where new pesticides such as organochlorines and nicotine accounted for 55%, and organic phosphorus, pyrethroids, and carbamates accounted for 17.5%, 15.0%, and 12.5%, respectively. Seventeen types of pesticides exceeded the standard to varying degrees, including 12 types(70.59%) with exceeding rates not higher than 5%, four types(23.53%) with exceeding rates in the range of 5%-10%, and one type(5.88%) with an exceeding rate higher than 10%. As revealed by the analysis results of the past five years, the pesticide residue pollution of Chinese medicinal materials showed a downward trend. Compared with the conditions at worst, organochlorines decreased by about 2/3 in detection rate and 47.23% in exceeding rate, carbamates by about 1/2 in detection rate and 10.78% in exceeding rate, organic phosphorus by 3/4 in detection rate and 7.22% in exceeding rate, pyrethroids by 1/2 in detection rate and 11.05% in exceeding rate, and other types by about 1/2 in detection rate but not exceeded the standard. In general, pesticide residues in Chinese medicinal materials and safety have been significantly improved. However, pesticide residues are still important factors affecting the quality and safety of Chinese medicinal materials. It is suggested to further improve the control standards of pesticide residues in Chinese medicinal materials, strengthen the monitoring of pesticides used in practical production, and promote the ecological planting mode to facilitate the high-quality development of the Chinese medicinal material industry.

ABSTRACT

The continuous cropping obstacle of Panax notoginseng is serious, and effective control measures are lacking. Soil disinfection with chloropicrin(CP) has been proven to be effective in reducing the obstacles to continuous cropping of other crops. In order to ascertain the effect of CP in the continuous cropping of P. notoginseng, this paper explored the influences of CP at different treatment concentrations(0,30,40,50 kg/Mu, 1 Mu≈667 m~2) on soil macro-element nutrients, soil enzyme activity, growth and development of P. notoginseng, and the accumulation of medicinal components. The results showed that CP fumigation significantly increased the content of total nitrogen, alkali-hydrolyzable nitrogen, ammonium nitrogen, nitrate nitrogen, and available phosphorus in the soil, but it had no significant effect on potassium content. The soil protease activity showed a trend of first increasing and then decreasing with the prolonging of the treatment time. Both the soil urease and acid phosphatase activities showed a trend of first decreasing and then increasing with the prolonging of the treatment time. The higher the CP treatment concentration was, the lower the urease and acid phosphatase activities would be in the soil. The protease activity was relatively high after CP40 treatment, which was better than CP30 and CP50 treatments in promoting the nitrogen-phosphorus-potassium accumulation in P. notoginseng. The seedling survival rates after CP0, CP30, CP40, and CP50 tratments in October were 0, 65.56%, 89.44%, and 83.33%, respectively. Compared with the CP30 and CP50 treatments, CP40 treatment significantly facilitated the growth and development of P. notoginseng, the increase in fresh and dry weights, and the accumulation of root saponins. In summary, CP40 treatment accelerates the increase in soil nitrogen and phosphorus nutrients and their accumulation in P. notoginseng, elevates the seedling survival rate of P. notoginseng, enhances the growth and development of P. notoginseng, and promotes the accumulation of medicinal components. CP40 treatment is therefore recommended in production.

ABSTRACT

1,2,3-trichloropropane (TCP) is an industrially synthesized aliphatic chlorinated hydrocarbon and an intermediate product in the industrial production of epichlorohydrin, which can be used as a precursor for the manufacture of soil fumigant and organic solvents. Due to its biological toxicity, environmental persistence and strong environmental migration ability, 1,2,3-TCP is listed as an emerging organochlorine pollutant in the environment and regulated by many international organizations. Currently, the degradation of 1,2,3-TCP and the remediation of 1,2,3-TCP-contaminated sites receive great attention, but the degradation mechanism of 1,2,3-TCP has not been summarized in depth. This article discussed the origin of 1,2,3-TCP, its environmental impact and ecological effects, and the physical and chemical degradation techniques. This was followed by summarizing the degradation mechanisms of 1,2,3-TCP (e.g., aerobic co-biodegradation, anaerobic biodegradation). Specially, the pathways and mechanisms of microbial biodegradation and transformation of 1,2,3-TCP in anoxic environments (e.g., groundwater) were thoroughly reviewed. The feasibility of using 1,2,3-TCP as an electron acceptor by organohalide-respiring bacteria under anoxic conditions was predicted based on thermodynamic analysis. Last but not least, in situ bioremediation of 1,2,3-TCP contaminated sites was summarized, and prospects for future research were discussed.

ABSTRACT

Chlorinated hydrocarbons (CAHs) threaten human health and the ecological environment due to their strong carcinogenic, teratogenic, mutagenic and heritable properties. Heterotrophic assimilation degradation can completely and effectively degrade CAHs, without secondary pollution. However, it is crucial to comprehensively understand the heterotrophic assimilation process of CAHs for its application. Therefore, we review here the characteristics and advantages of heterotrophic assimilation degradation of CAHs. Moreover, we systematically summarize current research status of heterotrophic assimilation of CAHs. Furthermore, we analyze bacterial genera and metabolism, key enzymes and characteristic genes involved in the metabolic process. Finally, we indicate existing problems of heterotrophic assimilation research and future research needs.

ABSTRACT

Abstract This study sought to investigate the association of exposure to organochlorine (OC) and non-persistent pesticides with hematological parameters in an agricultural population in Southern Brazil. A cross-sectional study was conducted with a random sample of 275 farm workers and their families in Farroupilha-RS. A questionnaire was used to collect information on sociodemographic and lifestyle factors, duration, frequency and type of pesticide used, among others. Blood samples were collected and analyzed for serum concentration of 24 OC pesticides and hematological parameters. Associations were explored through linear regression, controlling for confounders. Lifetime use of chemical classes other than organophosphates and dithiocarbamates were associated with decreased number of lymphocytes, while subjects sampled in the high pesticide use season showed higher number of erythrocytes and hemoglobin level. Detectable serum levels of many OC pesticides were associated with lower counts of white blood cells, particularly eosinophils. Although mostly null associations were observed between pesticide use and hematological parameters, findings may suggest that OC pesticides could lead to hematological alterations among agricultural workers.


Resumo O objetivo deste estudo foi investigar a associação entre a exposição a organoclorados (OC) e agrotóxicos não persistentes e os parâmetros hematológicos em uma população agrícola de Farroupilha-RS. Foi utilizado um questionário para coletar informações sobre fatores sociodemográficos e de estilo de vida, duração, frequência e tipo de pesticidas utilizados, entre outros. Amostras sanguíneas foram coletadas e analisadas quanto a concentração sérica de 24 pesticidas OC e parâmetros hematológicos. As associações foram exploradas através de regressão linear, controlando por confundidores. O uso cumulativo de classes químicas diferentes de organofosforados e ditiocarbamatos associou-se com diminuição do número de linfócitos enquanto indivíduos que tiveram suas coletas sanguíneas realizadas na estação de maior uso de agrotóxicos tinham contagem de eritrócitos e hemoglobina maiores. Níveis séricos de diversos pesticidas organoclorados foram associados com contagens mais baixas de células brancas do sangue, particularmente eosinófilos. Embora as associações com o uso de agrotóxicos tenham sido, em geral, nulas, os resultados podem sugerir que os pesticidas OCs poderiam levar a alterações hematológicas entre os trabalhadores agrícolas.

ABSTRACT

BACKGROUND: Evidence on associations between occupational diesel exhaust and gasoline exposure and colorectal cancer is limited. We aimed to assess the effect of workplace exposure to diesel exhaust and gasoline on the risk of colorectal cancer. METHODS: This caseecontrol study included 181,709 colon cancer and 109,227 rectal cancer cases diagnosed between 1961 and 2005 in Finland, Iceland, Norway, and Sweden. Cases and controls were identified from the Nordic Occupational Cancer Study cohort and matched for country, birth year, and sex. Diesel exhaust and gasoline exposure values were assigned by country-specific job-exposure matrices. Odds ratios and 95% confidence intervals were calculated by using conditional logistic regression models. The results were adjusted for physical strain at work and occupational exposure to benzene, formaldehyde, ionizing radiation, chlorinated hydrocarbons, chromium, and wood dust. RESULTS: Diesel exhaust exposure was associated with a small increase in the risk of rectal cancer (odds ratio 1/4 1.05, 95% confidence interval 1.02–1.08). Gasoline exposure was not associated with colorectal cancer risk. CONCLUSION: This study showed a small risk increase for rectal cancer after workplace diesel exhaust exposure. However, this finding could be due to chance, given the limitations of the study.

ABSTRACT

This study used gas chromatography-mass spectrometry (GC-MS) to detect the products formed during the contact of sodium hypochlorite (NaOCl) with bovine pulp and dentin. For analysis of the products formed in the volatile phase, 11 mg of bovine pulp tissue were placed in contact with 0.5%, 2.5% and 5.25% NaOCl until complete tissue dissolution occurred. The solid phase microextraction (SPME) fiber was exposed inside the container through the cover membrane and immediately injected into the GC-MS system. 30 mg of the of dentin were kept in contact with NaOCl, and then the SPME fiber was exposed inside the container through the cover membrane for adsorption of the products and injected into the GC-MS system. The same protocol was used for the aqueous phase. For analysis of the volatile compounds, the final solution was extracted using pure ethyl ether. The suspended particulate phase of the mixture was aspirated, and ether was separated from the aqueous phase of the solution. The ether containing the products that resulted from the chemical interaction of dentin and pulp with the NaOCl was filtered and then injected into the GC-MS system for analysis of the aqueous phase. The aqueous and volatile phases of both dentin and pulp showed the formation of chloroform, hexachloroethane, dichloromethylbenzene and benzaldehyde. In conclusion, organochlorine compounds are generated during the contact of dentin and pulp with NaOCl at concentrations of 0.5%, 2.5% and 5.25%.


Este estudo utilizou a cromatografia gasosa acoplada a espectrometria de massa (CG-MS) para detectar os produtos que se formaram durante o contato de hipoclorito de sódio (NaOCl) com polpa dental bovina e dentina. Para a análise dos produtos formados na fase volátil, 11 mg de polpa bovina foram colocados em contato com 0,5 % , 2,5 % e 5,25 % de NaOCl, até à dissolução completa dos tecidos. A fibra de microextração em fase sólida (SPME) era exposta dentro do recipiente através da membrana da tampa, por 15 minutos, para a adsorção dos produtos formados e imediatamente injetada no CG-MS para análise. Para a análise da dentina, 30 mg do de amostras foram mantidas em contacto com o NaOCl, por 15 min, e então a fibra de SPME era exposta no interior do recipiente através da membrana de cobertura para a adsorção dos produtos e injectado no sistema de GC-MS. O mesmo protocolo foi utilizado para a fase aquosa. Para a análise dos compostos voláteis, a solução final foi extraída com éter etílico puro. A fase de partículas em suspensão da mistura foi aspirada, e o éter foi separado da fase aquosa da solução. O éter contendo os produtos que resultaram da interacção química da dentina e polpa com hipoclorito de sódio foi filtrado e, em seguida, injectado no sistema GC-MS para análise da fase aquosa. As fases aquosas e voláteis de dentina e polpa mostraram a formação de clorofórmio, hexacloroetano, dichloromethylbenzene e benzaldeído. Compostos organoclorados são gerados durante o contacto da dentina e polpa com hipoclorito de sódio em concentrações de 0,5 % , 2,5 % e 5,25 %.

ABSTRACT

Bioremediation is one of the most effective ways to treat and dispose of chlorinated hydrocarbons, and methanotrophs are potentially useful to do so. Recent studies found that facultative methanotrophs can use compounds containing C-C bond as sources of carbon and energy, thus overcoming the limitation that obligate methanotrophsone uses only C1 compounds for this process. This is a unique metabolic approach that is becoming increasingly attractive in the field of contaminant biodegradation. Here, we summarized the bioremediation of chlorinated hydrocarbons by obligate and facultative methanotrophs. This process involves the degradation of various chlorinated hydrocarbons by diverse strains, including pure cultures and mixed cultures. We also compare the activity expression and catalytic properties of different types of methane monooxygenases in various substrates. We furthermore summarize the kinetic characteristics of the degradation of chlorinated hydrocarbons using the model strain Methylosinus trichosporium OB3b, and outline the degradation and potential of chlorinated hydrocarbons by facultative methanotrophs. Lastly, we discuss current problems and future research directions for degradation of chlorinated hydrocarbons by methanotrophs.

<p><b>OBJECTIVE</b>To establish a method for determination of chloropicrin in the air of workplace by gas chromatography with liquid absorption collection.</p><p><b>METHODS</b>Chloropicrin in sample air was collected by absolute ethyl alcohol in a porous glass plate absorption tube. Following direct sample loading, samples were separated by gas chromatography and detected with a micro-electron capture detector. The present method was compared with the national standard method.</p><p><b>RESULTS</b>The range of linearity was 0-3.0 µg/ml, and the regression equation was y = 3488.80x-57.84, with a correlation coefficient of 0.999 4. The detection limit was 0.003 µg/ml, with a minimum detectable concentration of 0.005 mg/m³ based on a 3.0 L sampling volume. The average recovery rate was 98.9%∼103.2%. The within-run precision was 0.96%-2.12%, and the between-run precision was 2.75%-4.59%. The sampling efficiency was 98.6%. Samples in porous glass plate absorption tube could be stored at 4 °C for at least 5 days. The result ratio of this method to the national standard method was 97.7%-108.0% .</p><p><b>CONCLUSION</b>The present method meets the requirements of "Guide for establishing occupational health standards-Part 4: Determination methods of air chemicals in workplace" (GBZ/T 210.4-2008), and is feasible for determination of chloropicrin in the air of workplace.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effect of xuebijing Injection (XI) on perioperative coagulation and inflammatory reaction in senile patients receiving total hip arthroplasty (THA).</p><p><b>METHODS</b>Totally eighty patients receiving THA at Luoyang Orthopedics Hospital, 65 to 85 years old, were randomly assigned to the control group (40 cases) and the treatment group (40 cases). All patients received routine perioperative therapies. Those in the treatment group received XI (adding 50 mL XI in 100 mL normal saline, 30 min each time). XI was continually injected after THA, twice daily for 3 successive days. Blood samples were harvested on the morning of the 2nd admission day (TO), immediately after operation (T1), on the morning of the 3rd day after operation (T3), and on the morning of the 5th day after operation (T4) to detect prothrombin time (PT), thrombin time (TT), activated partial thromboplastin time (APTT), levels of FIB and D-dimer (D-D), changes of white blood cell (WBC), neutrophils (N), C-reactive protein (CRP), erythrocyte sedimentation rate (ESR), and IL-6. Complications of surgery were compared between the two groups.</p><p><b>RESULTS</b>There was no statistical difference in operation time, intraoperative blood loss, and blood transfusion between the two groups (P >0.05). Compared with TO in the same group, WBC, N, CRP, ESR, IL-6, PT, TT, and D-D all increased in the control group at T1-T4 (P < 0.05); APTT increased at T1-T2 (P <0.05); FIB increased at T1-T3 (P <0.05). WBC, N, IL-6, PT, and D-D all increased in the treatment group at T1-T3 (P <0.05); CRP and ESR increased at T1-T4 (P < 0.05); TT increased at T1-T2 (P <0.05); APTT and FIB increased at T1 (P <0.05). Compared with the control group at the same time period, WBC, N, CRP, and IL-6 all decreased in the treatment group at T1-T4 (P <0.05), ESR decreased at T3-T4 (P <0.05); PT and TT decreased at T1-T3 (P <0.05); FIB and D-D decreased at T2-T4 (P<0.05). The occurrence of each complication was significantly lower in the treatment groups than in the control group.</p><p><b>CONCLUSION</b>XI could improve the perioperative high coagulation state of senile THA patients, inhibit inflammatory reactions, and reduce complications.</p>

ABSTRACT

OBJECTIVE: The aim of the study was quantify organochlorine compounds in women seeking for infertility treatment (n = 15) and in spontaneously pregnant ones (n = 21). MATERIALS AND METHODS: A questionnaire was applied regarding lifestyle, occupational and reproductive history. Blood samples were collected from both groups. RESULTS: From the pesticides studied, pp'DDE was detected in 100% of infertile women, at higher mean levels than in pregnant women (3.02 mcg/L vs. 0.88 mcg/L; p = 0.001; power of 69%), without correlation with the etiology of infertility. Levels of the polychlorinated biphenyls (PCBs) were low, with positive samples in 100% in the infertile women for PCBs 138, 153, 180, while in pregnant women, they were 85.7% for congeners 138 and 153. Only PCB180 showed significance, with frequency of 71.4% (p = 0.019). CONCLUSIONS: The risk factors for female infertility were: age, consumption of untreated water and of canned foods. Exposure to the most prevalent organochlorine compounds described in literature was confirmed in the study, indicating that pp'DDE may adversely influence female fertility.


OBJETIVO: O estudo teve como objetivo quantificar as substâncias organocloradas em mulheres buscando tratamento para infertilidade (n = 15) e que espontaneamente engravidaram (n = 21). MATERIAIS E MÉTODOS: Foi aplicado questionário considerando estilo de vida, história ocupacional e reprodutiva. Amostras de sangue foram obtidas em ambos os grupos. RESULTADOS: Dos pesticidas, pp'DDE foi detectado em 100% das inférteis, com níveis maiores que nas grávidas (3,02 mcg/L vs. 0,88 mcg/L; p = 0,001; poder 69%), sem correlação na etiologia da infertilidade. Os níveis de detecção das bifenilas policloradas (PCBs) foram baixos, com 100% de positividade das amostras nas inférteis para os PCBs 138, 153, 180, e de 85,7% nas grávidas para os congêneres 138 e 153. Apenas PCB180 mostrou significância na frequência de 71,4% (p = 0,019). CONCLUSÕES: Os fatores de risco para infertilidade feminina foram: idade, consumo de água não tratada e alimentos enlatados. A exposição aos organoclorados mais prevalentes descritos na literatura foi confirmada no estudo, indicando que pp'DDE pode influenciar adversamente a fertilidade feminina.

ABSTRACT

O presente trabalho objetivou apresentar os resultados da aplicação da análise de agrupamento (cluster) para a caracterização da exposição a organoclorados, através de variáveis relacionadas ao tempo e à dose de exposição. Características de 354 sujeitos de uma população exposta a resíduos de pesticidas organoclorados relacionadas ao tempo e à dose de exposição foram submetidas à análise de cluster para separá-las em subgrupos. Foi realizada a análise de cluster hierárquico. Para avaliar a precisão da classificação, foram comparadas a variabilidade intragrupo e a variabilidade inter-grupos através do teste de ANOVA para cada dimensão. A estratégia de agregação foi realizada pelo método de Ward. Para criação dos clusters, foram comparadas as variáveis associadas à exposição e às rotas de contaminação. As informações referentes às doses estimadas de ingesta do composto foram usadas para ponderar os valores de tempo de exposição a cada uma das rotas, de forma a obter valores proxy de intensidade de exposição. Os resultados indicaram 3 clusters: o cluster 1 (n = 45), com características de maior exposição; o cluster 2 (n = 103), de exposição intermediária, e o cluster 3 (n = 206), de menor exposição. As análises bivariadas realizadas com os grupos evidenciaram que são grupamentos com diferença estatisticamente significativa. Este estudo evidenciou a aplicabilidade da análise de cluster para categorizar populações expostas a organoclorados, e aponta para a relevância de estudos tipológicos que possam contribuir para uma melhor classificação de sujeitos expostos a agentes químicos, condição típica dos estudos de epidemiologia ambiental, para uma mais ampla compreensão dos aspectos etiológicos, preventivos e terapêuticos da contaminação.


This study aimed to show the results from a cluster analysis application in the characterization of population exposure to organochlorines through variables related to time and exposure dose. Characteristics of 354 subjects in a population exposed to organochlorine pesticides residues related to time and exposure dose were subjected to cluster analysis to separate them into subgroups. We performed hierarchical cluster analysis. To evaluate the classification accuracy, compared to intra-group and inter-group variability by ANOVA for each dimension. The aggregation strategy was accomplished by the method of Ward. It was, for the creation of clusters, variables associated with exposure and routes of contamination. The information on the estimated intake doses of compound were used to weight the values of exposure time at each of the routes, so as to obtain values proxy exposure intensity. The results showed three clusters: cluster 1 (n = 45), characteristics of greatest exposure, the cluster 2 (n = 103), intermediate exposure, and cluster 3 (n = 206), less exposure. The bivariate analyzes performed with groups that are groups showed a statistically significant difference. This study demonstrated the applicability of cluster analysis to categorize populations exposed to organochlorines and also points to the relevance of typological studies that may contribute to a better classification of subjects exposed to chemical agents, which is typical of environmental epidemiology studies to a wider understanding of etiological, preventive and therapeutic contamination.

ABSTRACT

Zebra fish were exposed to environmentally relevant concentration of pesticides and metals for a period of 14 days. The individual and the combined toxicity of pesticides and metals were studied. Damage caused to the DNA and induction of mutation in the GADD45beta gene was investigated in this study. The present investigation revealed that exposure of zebra fish to pesticides and metals induced DNA damage and also mutation in the GADD45beta gene. The pesticides treated group [III] showed significantly higher damage followed by metals treated group [II]. The DNA damage was comparatively less in the pesticides and metals exposed group [IV]. GADD45beta gene which is considered to be one of the most important gene involved in G phase arrest, has been found to be mutated in zebra fish when exposed to metals and pesticides. The comparison of amplified gene sequence of GADD45beta gene with the sequence of the zebra fish retrieved from the NCBI confirmed the mutation in the gene sequence of zebra fish treated with pesticides and metals. The induction of DNA damage in fish due to contaminants may lead to mutations, which contribute towards the multistage carcinogenesis process

There are substantial variations of relative risks (RR) in smoking-related mortality by country and time. We hypothesized the RRs in smoking-related mortality might differ depending on serum concentrations of persistent organic pollutants (POPs). We evaluated the associations of cigarette smoking with total mortality in 610 elderly (aged > or = 70 yr) (702 elderly for organochlorine pesticides [OCPs]) after stratification by serum concentration of POPs, in the National Health and Nutrition Examination Survey (NHANES) 1999-2004 followed through 2006. Summary measures of POPs subclasses showed significant or marginally significant interaction with cigarette smoking on the risk of total mortality. P values for interaction were 0.069 for polychlorinated dibenzo-p-dioxins (PCDDs), 0.008 for polychlorinated biphenyls (PCBs), and 0.024 for OCPs. The effect of smoking on total mortality showed different patterns according to the serum concentration of some POPs. Former or current smokers had 1.4 to 2.9 times higher mortality rates compared with never smokers among participants with higher serum concentrations of POPs (2nd or 3rd tertiles). However, when the level of PCBs or OCPs were low (1st tertile), there were little positive associations between smoking and mortality. Our study suggests that the background exposure to several POPs may be related to variability in smoking-related total mortality.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the correlation between pro coagulation factors and anti-coagulation factors synthesized by the liver, and the correlation between fibrin degradation products (FDP) and D-dimer (D-D) concentration and coagulation proteins synthesized by extra-hepatic tissues, in different liver diseases; to explore the relationship between coagulation and bleeding in hepatic diseases.</p><p><b>METHODS</b>Chronic hepatitis B (CHB) patients, CHB-related liver cirrhosis patients, CHB-related liver failure patients and healthy (normal) controls were selected for study and provided blood samples for analysis. The activity of coagulation factors (F) II, V, VII, VIII, IX, X, XI, and XII was detected using the one-stage clotting method. Coagulogram analysis, including activated partial thromboplastin time (APTT), thrombin time (TT), and prothrombin time (PT), was conducted by the solidification method. Antithrombin III (AT-III) and protein C (PC) activities were measured by chromogenic substrate assay. FDP concentration was detected using immunoturbidimetry. Tissue factor pathway inhibitor (TFPI), thrombomodulin (TM), von Willebrand factor (vWF), and tissue factor (TF) concentrations were measured by enzyme-linked immunosorbent assay (ELISA).</p><p><b>RESULTS</b>With the exception of FVIII, coagulation factors and anticoagulant proteins synthesized by the liver were decreased and the coagulogram was extended for all patients. Likewise, the FDP and D-D concentrations were increased in blood. CHB patients, however, presented with increased levels of FVIII, TFPI, TM, vWF, and TF. Pairwise comparison indicated statistical differences existed among CHB, CHB-related liver cirrhosis, and liver failure patients: TFPI: 239.3+/-206.4, 315.0+/-258.6, and 319.5+/-298.1 -- higher than normal control: 104.0+/-87.1, F = 5.453, P less than 0.05; vWF: 70.3+/-29.5, 105.5+/-58.0, and 179.3+/-61.7 -- higher than normal control: 21.9+/-7.2, F = 20.104, P less than 0.05; TF: 85.9+/-85.7, 234.2+/-202.9, and 344.7+/-214.6 -- higher than normal control: 12.8+/-8.1, F = 8.619, P less than 0.05; FVIII: 157.2+/-53.4, 206.9+/-86.9, and 335.7+/-117.7 -- higher than normal control: 105.5+/-46.2, F = 13.418, P less than 0.05.</p><p><b>CONCLUSION</b>In parallel to the progression of liver diseases, pro coagulation and anti-coagulation elements synthesized by the liver were reduced. In contrast, fibrinolysis activity was enhanced, which is expected to lead to an imbalance between blood clotting and anti-clotting factors. This may be an important cause for the bleeding that occurs in end-stage liver disease. Expressions of TFPI, TM, vWF, and TF significantly change in the early stage of liver diseases, as compared to normal (healthy) levels, and may represent a sensitive indicator of vascular injury.</p>

ABSTRACT

OBJECTIVES: Chronic inflammation is now thought to play a key pathogenetic role in the associations of obesity with insulin resistance and diabetes. Based on our recent findings on persistent organic pollutants (POPs) including the lack of an association between obesity and either insulin resistance or diabetes prevalence among subjects with very low concentrations of POPs, we hypothesized that POP concentrations may be associated with inflammation and modify the associations between inflammation and insulin resistance in non-diabetic subjects. METHODS: Cross-sectional associations among serum POPs, C-reactive protein (CRP), and homeostasis model assessment of insulin resistance (HOMA-IR) were investigated in 748 non-diabetic participants aged > or =20 years. Nineteen types of POPs in 5 subclasses were selected because the POPs were detectable in > or =60% of the participants. RESULTS: Among the five subclasses of POPs, only organochlorine (OC) pesticides showed positive associations with CRP concentrations, while polychlorinated biphenyls (PCBs) showed inverse associations with CRP concentrations. There were statistically significant interactions between CRP and OC pesticides and between CRP and PCBs, in estimating HOMA-IR (P for interaction <0.01 and <0.01, respectively). CRP was not associated with HOMA-IR among subjects with low concentrations of OC pesticides or PCBs, while CRP was strongly associated with HOMA-IR among subjects with high concentrations of these POPs. CONCLUSIONS: In the current study, OC pesticides were associated with increased levels of CRP, a marker of inflammation, and both OC pesticides and PCBs may also modify the associations between CRP and insulin resistance.

ABSTRACT

Objetivo Evaluar la presencia de pesticidas organoclorados (OCPs) en leche cruda bovina procedente de fincas de tradición algodonera del municipio de San Pedro, Sucre. Materiales y métodos Se tomaron 48 muestras de leche procedente de tres fincas ganaderas, dos de ellas con tradición en la suplementación alimenticia con semillas y soca de algodón, y se les determinó la concentración de OCPs por cromatografía gaseosa empleando un detector de microcaptura de electrones (µECD). Resultados El 100 por ciento de las muestras presentaron algún tipo de OCPs, y las concentraciones más altas correspondieron a los pesticidas Endrin, DDT y Aldrin, con 0,62, 0,59 y 0,52 µg/mL, respectivamente. Las fincas que suplementaron con soca de algodón tuvieron una mayor presencia y concentración de OCPs en leche que las muestras provenientes de la finca que no realizaba esta práctica. Conclusión A pesar que la mayoría de los plaguicidas organoclorados están prohibidos en el país desde 1986, aún se presentan residuos en las muestras de leche cruda analizadas provenientes de fincas con tradición en el cultivo de algodón, generando un riesgo de salud pública a los consumidores.


Objective Ascertaining the presence of organochlorine pesticides (OCPs) in raw milk from cows grazing on traditional cotton farms in San Pedro, Sucre, Colombia. Materials and methods 48 samples were taken from three cattle farms; two of them used cotton seed and soca dietary supplement.OCP concentration was determined by gas chromatography (GC) using an electron capturedetector (µ-ECD). Results 100 percent of the samples had some type of OCPs and Endrin, DDT and Aldrin pesticides hadthe highest concentrations (0.62, 0.59 and 0.52 µg/mL, respectively). Farms supplementing feed with soca cotton had the greatest OCP presence and concentration in milk samples compared to samples from farms which did not engage in this practice. Conclusion Although most OCPs have been banned in Colombia since 1986, residues are still being presented in raw milk samples tested from cottongrowing farms, thereby creating a public health risk for consumers.

Los productos en desarrollo y los niños son particularmente vulnerables a los tóxicos ambientales. Nuestro objetivo es determinar los niveles de plaguicidas organoclorados (HCB, ß-HCH, pp'DDT, op'DDT y pp'DDE) en lípidos séricos de madres de niños con criptorquidia y comparar los niveles con un grupo control de madres de niños con testículos descendidos. El grupo de los casos fue constituido por recién nacidos con diagnóstico de criptorquidia (n=41). El grupo control (n=41) se conformó por niños con testículos descendidos. A las madres de ambos grupos se les determinaron los niveles de plaguicidas organoclorados. La criptorquidia fue diagnosticada al nacimiento por neonatólogo. Residuos de plaguicidas organoclorados fueron encontrados en lípidos séricos de ambos grupos. Los niveles de la mediana (mgkg-1 en base lipídica) fueron mayores para los metabolitos pp'DDT (0.464 vs. 0.269) y ß-HCH (0.263 vs. 0.192) en el grupo con criptorquidia comparado con el grupo control. (p<0.01). Los niveles de los metabolitos pp'DDT y ß-HCH son mayores entre las madres de los recién nacidos con criptorquidia. Es posible que sustancias con efectos antiandrogénicos puedan producir disrupción endocrina y criptorquidia durante el desarrollo fetal.


Fetuses and children are more susceptible to the effects of environmental toxins. The objective of this article is to determine the levels of organochlorine pesticides (HCB, ß-HCH, pp'DDT, op'DDT and pp'DDE) in the serum lipids of mothers of newborns with cryptorchidism and compare the levels to a control group of mothers of newborns with descended testicles. The cases were composed of newborns with cryptorchidism (n=41), and the controls (n=41) newborns with descended testicles. Blood samples from both groups of mothers were used to determine the organochlorine pesticide levels. Cryptorchidism was diagnosed at birth by a neonatologist. The results showed that the organochlorine pesticide residues were found in the serum lipids of both groups of mothers. The median serum lipid levels (mgkg-1 lipid-based) were statistically higher for the metabolites pp'DDT (0.464 vs. 0.269) and ß-HCH (0.263 vs. 0.192) in the cryptorchidism group compared to the control group (p<0.01). It could be concluded that the levels of the metabolites pp'DDT and ß-HCH are higher among mothers of newborns with cryptorchidism. It is possible that substances with anti-androgenic effects could produce endocrine disruption, such as cryptorchidism, during fetal development.
